UC Berkeley researchers have identified a novel therapeutic approach for obesity and diabetes using a molecular compound called TOFA (5-tetradecyloxy-2-furoic acid). Unlike GLP-1 medications that suppress appetite, this treatment “boost[s] the body’s metabolic rate” by increasing energy expenditure.
Key Findings
The compound works through dual mechanisms: blocking lipid production while activating cellular receptors that enable fat burning. In mouse studies, TOFA caused cells to burn approximately 18% more energy without changes in activity levels or body temperature. Notably, obese mice experienced fat loss while preserving lean muscle massโa significant advantage over appetite-suppressing drugs.
The researchers discovered TOFA functions as both an ACC inhibitor and a PPARฮฑ/PPARฮด activator, creating what researcher Justin Lee describes as “a coordinated metabolic response” rather than simple lipid blocking.
Clinical Potential
Testing showed TOFA combined with GLP-1 drugs produced superior results compared to either treatment alone, suggesting complementary rather than competitive mechanisms.
Important caveat: The research remains preliminary, conducted only in animal models. Human safety and efficacy testing has not yet begun.
